NT-I7 restores T cell levels and enhances antitumor efficacy in head and neck cancer models Strong synergistic effects observed when combined with cisplatin and immune checkpoint inhibitors April 25, 2025 — ROCKVILLE, MD : NeoImmuneTech, Inc. (NIT or “NeoImmuneTech”), a T cell-focused therapeutics company, announced today that it will present preclinical data on NT-I7 (efineptakin alfa) at the American Association for Cancer Research (AACR) Annual Meeting 2025, to be held from April 25 to 30 in Chicago, USA. The study demonstrates NT-I7’s ability to restore lymphocyte levels and enhance anti-important effects in head and neck squamous cell carcinoma (HNSCC) models. This study, conducted in collaboration with the laboratories of Dr. Daniel Johnson and Dr. Matthew Spitzer at the University of California, San Francisco (UCSF), focused on the immune synergy of NT-I7 when combined with cisplatin, commonly used in standard of care for HNSCC, and immune checkpoint inhibitors(anti-PD-1). In HNSCC tumor-bearing mouse models, NT-I7 significantly restored the reduced populations of CD4+, CD8+ T cells, and NKT cells, without affecting tumor-infiltrating regulatory T cells (Tregs). In the MOC22 tumor model, NT-I7 monotherapy showed strong antitumor efficacy, and in the MOC1 model, a combination of NT-I7 and cisplatin yielded significant tumor suppression. Additionally, immune profiling using CyTOF (Cytometry by Time of Flight) revealed that NT-I7 promoted the expansion of the CD8+ Tpex (precursor exhausted T cells) subpopulation responsive to checkpoint inhibitors, supporting its potential as a mechanism-based combination therapy. NIT noted that the increase in Tpex is consistent across multiple clinical studies with NT-I7.
