OBI Pharma (4174.TWO)’s independent work based on a TROP2-targeted Antibody-Drug Conjugate (ADC) anti-cancer asset, OBI-992, is published on the cover of the internationally renowned journal “Molecular Cancer Therapeutics” (MCT), with the article designated as a First Disclosure recommended publication. Molecular Cancer Therapeutics, a journal of the American Association for Cancer Research (AACR), specializes in preclinical cancer drug development and is known by its rigorous and high standards. OBI Pharma’s paper, titled “OBI-992, a novel TROP2-targeted antibody-drug conjugate, demonstrates antitumor activity in multiple cancer models ” has been designated as a First Disclosure publication. According to MCT’s definition of “First Disclosure,” this classification signifies more than just the initial publication of the drug’s structure and research. The designation indicates that expert reviewers have meticulously examined the preclinical drug activity, translational potential, and scientific transparency. After comprehensive evaluation, the reviewers determined that OBI-992 demonstrates substantial clinical development potential, thereby selecting it as First Disclosure recommendation for publication. Dr. Wan-Fen Li, the first author of the paper, the Development Champion of the TROP2 ADC program at OBI, explained that OBI-992 is a novel ADC targeting TROP2. The antibody is linked with a potent TOP1 (topoisomerase I) inhibitor via a linker with remarkable hydrophilic properties. It allows OBI-992 to maintain stability in the bloodstream. Upon binding and internalizing with TROP2 on cancer cell surfaces, it releases the TOP1 inhibitor to effectively kill cancer cells. Notably, the ADC demonstrates a compelling “bystander effect” against TROP2-negative cancer cells, killing neighboring cancer cells and transcending traditional targeted therapeutic limitations. Dr. Wan-Fen Li emphasized that diverse animal models have demonstrated OBI-992’s robust anti-tumor activity, outstanding pharmacokinetic performance, and excellent safety profile. When combined with commonly used anti-cancer treatments such as PARP inhibitors or immune therapies, the combinations exhibited enhanced anti-tumor activity. These findings validate OBI-992 as an innovative ADC with the potential to become a best-in-class therapeutic asset. They also highlight its promising prospect for future combination therapies. Dr. Heidi Wang, CEO of OBI Pharma, noted that OBI-992 entered clinical trials in June, 2024. It was also shortlisted among the top eight “Most Promising Clinical Candidates” by the prestigious World ADC Awards for 2024.
