SAN FRANCISCO, U.S., SUZHOU, China May 15, 2025 – Innovent Biologics, Inc. (“Innovent”) (HKEX: 01801), a world-class biopharmaceutical company that develops, manufactures and commercializes high-quality medicines for the treatment of oncologic, autoimmune, cardiovascular and metabolic, ophthalmologic and other major diseases, today announced that preclinical data on its novel bispecific antibody, IBI3002, which targets both IL-4Rα and TSLP pathways simultaneously for potential treatment of asthma, will be presented at the American Thoracic Society (ATS) 2025 International Conference. The conference will be held in San Francisco, California from May 16-21, 2025. Research highlight: Session: D21 – Immunological Insights in Lung Inflammation and Repair Topic: Preclinical Characterization of IBI3002, an Anti-IL-4Rα and Anti-TSLP Bispecific Antibody That Potently Dampens Inflammatory Response and Alleviates Asthma in Mice Abstract Number: 801 Presentation Format: Poster Presentation Time: Wednesday, May 21, 2025, 8:15 AM - 10:15 AM PST Presenter: Yao Xiong, Associated Director, In-vivo Pharmacology Preclinical data have shown that IBI3002 effectively targets and inhibits the IL-4Rα and TSLP pathways, and its TSLP inhibitory activity is stronger than that of Tezepelumab, a benchmark monoclonal antibody approved for asthma. In vitro, IBI3002 robustly inhibits the release of inflammatory cytokines from immune cells, and its activity is significantly better than that of Dupilumab (anti-IL-4R) and Tezepelumab (anti-TSLP) as single target blockade. In multiple transgenic mouse models, IBI3002 demonstrates superior efficacy in reducing airway inflammation and immune cell infiltration, in comparison with the same doses of Dupilumab, Tezepelumab and a similar bispecific antibody benchmark, indicating the combinatorial and synergistic effects of dual-targeting. Besides, the molecule has demonstrated a good safety profile in monkey toxicology experiments. Collectively, this dual-targeting mechanism may lead to significant advances in the treatment of moderate-to-severe asthma and also has potential in other type 2 inflammatory diseases. Dr. Huizhong Xiong, Senior Director of Immunology at Innovent, commented: “based on our understanding of the disease biology, targeting multiple inflammatory pathways simultaneously can potentially provide superior efficacy and synergy over single target blockade in respiratory disease and other conditions. Despite of an increasing number of innovative drug candidates entering clinics recently, there are still great unmet medical needs due to large patient population and disease complexity and heterogeneity. Our preclinical results have shown that IBI3002, a first-in-class IL-4Rα/TSLP bispecific antibody with a unique mechanism, may lead to breakthrough therapeutic advances for asthma and the other inflammatory diseases. IBI3002 is currently being evaluated in a Phase 1 clinical trial and we have observed encouraging preliminary results. We look forward to continuing to advance the development of this drug candidate to gain a deeper understanding of its potential in different diseases and patient populations, and to explore it in more indications.” About IBI3002 IBI3002 is a global first-in-class bispecific antibody simultaneously targeting cell surface IL-4Rα and the alarmin cytokine TSLP. With its high-efficiency dual-blocking mechanism, IBI3002 has demonstrated superiority over marketed monoclonal antibodies to the respective targets in in vitro assays. By concurrently inhibiting both IL-4Rα and TSLP pathways, IBI3002 shows promising potential for treating various inflammatory disorders including asthma. IL-4 functions as a central mediator of type 2 lymphocyte differentiation, inducing production of type 2-associated cytokines (IL-5, IL-9, IL-13) and chemokines like thymus and activation-regulated chemokine (TARC/CCL17) and eotaxin-3. IL-4 regulates B-cell isotype class switching to produce IgE and promotes eosinophil recruitment through IL-5. While IL-13 shares some inflammatory functions with IL-4, it uniquely mediates goblet cell hyperplasia, mucus production, smooth muscle contractility, and airway hyperresponsiveness. Together, IL-4 and IL-13 are critical drivers of type 2 inflammation. TSLP is an epithelial cell-derived alarmin cytokine that occupies an upstream position in the inflammatory cascade, promoting type 2 inflammation and Th2 cell differentiation and maturation. Importantly, TSLP is also implicated in triggering non-type 2 inflammatory pathways in asthma, making it a valuable complementary target to IL-4Rα blockade. IBI3002 is currently being evaluated in a global Phase 1 clinical trial, and about to enter multiple-ascending dose (MAD) stage in the near term.
