6 August 2025 Highlights • Successful pre-clinical trial assessing efficacy of TriGlytza® in animal model of Type 2 Diabetes Mellitus (T2DM), reproducing and adding to previous data. • TriGlytza® efficacy endpoints for HbA1c % reduction were achieved over the 1 month of treatment with superiority to Metformin, the primary care in diabetes. • This pre-clinical data reinforces the potential efficacy of TriGlytza® in the treatment of T2DM. Sydney, Australia, 5th August 2025: Myopharm Limited, is pleased to announce positive significant efficacy data on the key endpoint of HbA1c % in its pre- clinical trial of TriGlytza® for the treatment of Type 2 Diabetes Mellitus (T2DM). TriGlytza, a combination of Metformin, Celecoxib, and valsartan, was previously shown to have activity in an animal model of T2DM, a diet-induced obesity (DIO) mouse model of insulin resistance, reducing non-fasting and fasting blood sugars, compared to Metformin the primary care of diabetes in Australia. Myopharm’s study commenced in June 2025 and explored TriGlytza® in the T2DM DIO mouse model compared to Metformin on the key endpoint HbA1c %. HbA1c % is a measure of long-term blood sugar levels, and is the key tool for diagnosing and monitoring diabetes, as it indicates how well blood sugar is controlled over time. Blood sugar tests measurement levels at a specific moment. HbA1c % is the primary endpoint in our TriGlytza phase II trial and used for registration of diabetes drugs in patients. Our goal: is to reduce metabolic diseases that millions of people worldwide are at risk of. Diabetes is the fastest growing condition globally with approximately 830 million adults around the world living with diabetes. World Health Organisation Results: The TriGlytza® pre-clinical trial successfully demonstrated statistically significant superiority of TriGlytza® in reducing HbA1c % over Metformin over the 1-month of treatment, meeting the key efficacy endpoint of superior HbA1c% reduction in the trial compared to Metformin, Celecoxib and Valsartan monotherapy and other controls. After 1-month of treatment mice treated with TriGlytza® achieved a mean (average) reduction in HbA1c % levels of 1.3 units, compared with a 0.3-unit reduction with Metformin, and no change in control mice receiving Valsartan, or Celecoxib, or just high fat diet, representing a p value of <0.05 off TriGlytza® vs Metformin. Other endpoints of the study of reducing non fasting glucose were met over 1-month with TriGlytza® demonstrating statistically significant superiority over Metformin reproducing the previous study data. Additionally, TriGlytza® showed a non-increase in weight, similar to Metformin, and superiority to Celecoxib and Valsartan monotherapy and the control mice on high fat diet which all gained weight. Professor Mark Cooper, AO, Professor of Diabetes, and head of the Department of Diabetes within the School of Translational medicine, and advisor to Myopharm, said: “These are promising results, and support the Phase II clinical development of TriGlytza® in T2DM seeking to help patients better manage their disease. Dr George Tachas, Myopharm CSO said “Full results from the TriGlytza® pre-clinical study are anticipated in 2025 with data to be presented at a conference, and we continue to explore intellectual property expansion opportunities”. Karinza Phoenix, CEO and Managing Director of Myopharm Limited, added: “These positive results reflect the transformative potential of TriGlytza® and position us well to proceed with our preparations for our Phase II clinical activities.”
