MEDIA RELEASE 18 DECEMBER 2023 Radiopharm’s 68Ga-Trivehexin (RAD 301) featured in prestigious peer- reviewed journal RSC Medicinal Chemistry • Study by Johannes Notni and colleagues confirms the molecular structure of 68Ga-Trivehexin (RAD 301) as optimal for pancreatic cancer imaging. • These data support the high affinity and selectivity of RAD 301 for αvβ6-integrin, while maintaining a low non-specific (off target) binding. Sydney, Australia – 18 December 2023 – Radiopharm Theranostics (ASX:RAD, “Radiopharm” or the “Company”), a developer of a world-class platform of radiopharmaceutical products for both diagnostic and therapeutic uses, is pleased to announce the recent publication of a study conducted by Johannes Notni and colleagues from Technical University of Munich and TRIMT GmbH, in the prestigious journal RSC Medicinal Chemistry, published by the Royal Society of Chemistry (RSC). The paper entitled “Complexity of αvβ6-integrin targeting RGD peptide trimers: emergence of non- specific binding by synergistic interaction” 1, describes the optimization of cancer-targeting molecules, culminating in the amino acid structure of 68Ga-Trivehexin (RAD 301) for tumor imaging. The paper has been highlighted as the cover story of the December issue of RSC Medicinal Chemistry. RAD 301 is a peptide-based molecule that targets αvβ6-integrin, a cellular marker for tumor invasion and metastatic growth, the expression of which correlates with decreased survival in several carcinomas, particularly pancreatic. The αvβ6-integrin receptor is found in high density on most pancreatic carcinoma cells, making it an attractive diagnostic and therapeutic target in Pancreatic Ductal Adenocarcinoma (PDAC) and Head-and-Neck Squamous Cell Carcinoma (HNSCC)2. This project resulted in the original discovery of Trivehexin, which quickly produced promising first- in-human PET imaging results based on low non-specific uptake and a high affinity and selectivity for αvβ6-integrin. The findings provide a deeper understanding of the technology’s potential role in advancing diagnostic imaging. RSC Medicinal Chemistry publishes significant research in medicinal chemistry and related drug discovery science. The research articles published in the journal must show a breakthrough or significant advance on previously published work, or bring new thinking or results that will have a strong impact in their field. 68Ga-Trivehexin is currently in Phase 1, as a PET imaging agent at the Montefiore Medical Center, Albert Einstein College of Medicine, NY, USA. The study will assess the safety, radiation dosimetry and imaging characteristics of RAD 301 in patients with advanced PDAC (pancreatic cancer). In May 2023, the FDA granted Radiopharm with an Orphan Drug Designation (ODD) for RAD 301 in pancreatic cancer. 1 Quigley NQ, Richter F, Kossatz S, Notni J. RSC Med. MEDIA RELEASE 18 DECEMBER 2023 Radiopharm Scientific Advisory Board member and co-inventor of the 68Ga-Trivehexin technology Dr. Johannes Notni said: "The optimization of the Trivehexin drug structure described in this article ultimately paved the way for a broad clinical application of 68Ga-Trivehexin in PET diagnostics. Our preclinical data on pharmacokinetics and PET image quality are compelling evidence for the potential of 68Ga-Trivehexin to make a difference in the diagnosis and ultimately the treatment of cancer and fibrotic diseases.” About Radiopharm Theranostics Radiopharm Theranostics is a clinical stage radiotherapeutics company developing a world-class platform of innovative radiopharmaceutical products for diagnostic and therapeutic applications in areas of high unmet medical need.
