In the paper titled, “CD22 modulation alleviates amyloid β-induced neuroinflammation”, the authors reported data unveiling the dual mechanism of action of Suciraslimab, a c linical-stage anti -human CD22 antibody, in addressing Alzheimer ’s disease . The study demonstrates that Suciraslimab not only alleviates neuroinflammation but also promotes the clearance of amyloid -beta (A β), a hallmark of Alzheimer ’s pathology. This dual action positions Suciraslimab as a potential promising therapeutic candidate for neurodegenerative diseases, particularly Alzheimer’s disease. Neuroinflammation is a critical factor in the progression of neurodegenerative diseases, including Alzheimer’s disease, where plasma soluble CD22 (sCD22) levels are notably elevated in Alzheimer’s disease patients. This article reported that sCD22 promotes neuroinflammation both in vivo and in vitro. The study reveals that CD22 interacts with amyloid-beta (Aβ), and Suciraslimab enhances the internalization of the CD22-Aβ complex in microglia, the key immune cells responsible for regulating neuroinflammation and a major contributor to neurodegenerative processes. These findings underscore the significance of sCD22 in driving neuroinflammation and demonstrate the dual therapeutic potential of S uciraslimab by targeting CD22: mitigating A β-induced inflammation and enhancing Aβ phagocytosis.
