Revolutionising Diabetes with Triglytza® 27 August 2026 SHAREHOLDER MYOPHARM REPORTS PRELIMINARY FINDINGS FROM TRIGLYTZA® TYPE 1 DIABETES PILOT STUDY Sydney, NSW, Australia —Myopharm Limited (Myopharm or the Company), a biopharmaceutical company developing novel therapies targeting diabetes and metabolic health, today announced preliminary findings from a pre -clinical pilot study evaluating TriGlytza® in a chemically induced model of insulin-deficient, type 1 diabetes (T1D). The study provides early evidence that TriGlytza treatment was associated with improved glucose responsiveness and numerical reductions in selected circulating immune -cell populations. The findings support further investigation of TriGlytza® in T1D models designed to assess β-cell preservation and immune regulation. Study model The study used streptozotocin (STZ) to produce insulin -deficient hyperglycaemia in male C57BL/6 mice. STZ is a pancreatic β-cell toxin that is commonly used in experimental diabetes research to produce substantial β-cell injury and loss of endogenous insulin production. Key findings 25% reduction in post-dose glucose After 15 days of treatment, TriGlytza® treated animals demonstrated a statistically significant 25% reduction in non-fasting blood glucose measured four hours after morning dosing, compared with STZ-vehicle animals. Evidence of glucose responsiveness At the end of the study, HOMA analysis (a blood test calculation used to estimate insulin resistance and pancreatic β-cell function from fasting glucose and insulin levels) indicated that 4 of 8 (50%) TriGlytza-treated animals were glucose responsive, compared with 3 of 10 (30%) STZ-vehicle animals. Forward-looking statements: This announcement may contain forward-looking statements regarding the RESILIENCE-2a clinical trial, including its expected design, timing and outcomes. Forward-looking statements are not guarantees of future performance and are subject to known and unknown risks and uncertainties that may cause actual results to differ materially from those expressed or implied. Myopharm does not undertake any obligation to update or revise any forward-looking statements, except as required by law. Numerical reduction in circulating immune cells The study showed a 38.8% reduction in circulating Neutrophils and a 50.8% reduction in circulating Monocytes. Neutrophils and Monocytes have been shown to regulate the autoimmune response in T1D by acting as early innate responders that infiltrate the pancreas, release cytotoxic mediators and activate adaptive autoreactive T cells that attack insulin - producing cells and drives T1D. Study interpretation Although the findings should be considered preliminary and hypothesis-generating, it supports further investigation of TriGlytza’s potential immunomodulatory and β-cell-preserving effects in Type 1 diabetes (T1D). Myopharm Executive Chair and CEO, Karinza Phoenix, said: “These preliminary findings provide an encouraging signal that TriGlytza ® may influence glucose responsiveness and selected immune-cell populations even in a model characterised by severe β-cell injury, providing an important foundation for TriGlytza into a new therapeutic area, Type 1 Diabetes. The next phase of our program will focus on expanding our research into other pre-clinical models of T1D and investigating the role of TriGlytza® as an immune and β-cell centric modulator in T1D.” About TriGlytza® TriGlytza® is an investigational fixed-dose combination of metformin, celecoxib and valsartan being developed by Myopharm. The programme is based on the hypothesis that simultaneously targeting metabolic, inflammatory and renin -angiotensin pathways may address multiple biological processes involved in diabetes progression. Myopharm will provide further updates as material clinical and operational milestones are achieved.
