Crescendo Biologics announces clinical biodistribution data of Humabody® bispecific CB307 at ESMO 2024 ▪ CB307 shows highly selective tumour targeting ▪ First clinical demonstration of the superior biodistribution of a Humabody® VH therapeutic within immuno-oncology Cambridge, UK, 14 September 2024 – Crescendo Biologics Ltd (Crescendo), a clinical stage, immuno- oncology company developing novel, targeted T cell enhancing therapeutics, today announced the presentation of a poster at the 2024 European Society for Medical Oncology (ESMO) Congress, taking place in Barcelona from 13-17 September. The poster was co-authored with clinicians and scientists from University of Groningen, Netherlands. The data confirms targeted biodistribution of the Company’s lead programme CB307, a ‘1st and only’ in class CD137 x PSMA bispecific for end-stage prostate cancer patients. As part of the ongoing Phase 1b POTENTIA trial, a clinical sub-study was conducted to evaluate the biodistribution of CB307, where CB307 was labelled with the radioactive tracer, Zirconium-89. The results show that CB307 successfully localised to the tumour with negligible uptake in PSMA-positive normal tissues, lymph nodes, bone marrow and salivary glands, aligning with the intended biodistribution profile. Theodora Harold, CEO at Crescendo Biologics, commented: “CB307’s selective and targeted tumour uptake in the clinic corroborates the superior tumour penetration and accumulation data we have seen preclinically when compared to traditional antibodies. Together with CB307’s excellent safety profile, these clinical results further validate our Humabody® VH platform's potential in the development of precision therapeutics.” Presentation Number 667P: Phase I study to assess biodistribution of CB307, a trispecific Humabody targeting CD137, prostate-specific membrane antigen, and human serum albumin with 89Zr-CB307 PET The poster is presented by Dr. Daan G. Knapen, medical oncologist at University Medical Center Groningen, on Saturday 14 September, and will be made available on the Publications section of the company’s website following their presentation at the meeting. -Ends- For more information, please contact: Crescendo Biologics ICR Consilium Theodora Harold, CEO + 44 (0) 12 2349 7140 [email protected] [email protected] Mary-Jane Elliott, David Daley, Sukaina Virji [email protected] About CB307 CB307 is Crescendo’s lead clinical candidate, a first-in-class, half-life extended, CD137 x PSMA bispecific Humabody®, designed to deliver a safer and more durable anti-cancer T cell response. CB307 conditionally activates only tumour-specific T cells, exclusively within the tumour microenvironment using the CD137 co-stimulatory mechanism. Its unique format enables potent, tumour-specific killing, while avoiding systemic toxicity and can be applied to a broad range of PSMA+ cancer indications to address a large unmet medical need. Clinical development of CB307 is on track with the Phase 1b POTENTIA trial ongoing in adult patients with PSMA+ metastatic castration-resistant prostate cancer (mCRPC). POTENTIA (NCT04839991) is an open- label, dose escalation and cohort expansion study to assess the safety, tolerability and preliminary efficacy of CB307 as a monotherapy and in combination with pembrolizumab (KEYTRUDA®).
